ABSTRACT

ObjectiveTo observe the effect of polysaccharides from root, stem, leaf and fruit of Schisandra chinensis on exercise endurance in the aging mice induced by D-galactose. MethodMale ICR mice were randomly assigned into six groups: blank control group, model group, root polysaccharide group, stem polysaccharide group, leaf polysaccharide group and fruit polysaccharide group. The mice were administrated with distilled water or root, stem, leaf and fruit polysaccharide (total sugar content of 35 mg·kg-1) by gavage. Thirty minutes after the administration, the blank control group was subcutaneously injected with normal saline, and the other groups with D-galactose (300 mg·kg-1), once daily for 6 weeks. The anti-fatigue effects were evaluated by rotarod test, forelimb grip strength test, and weight-loaded swimming test. The fatigue and oxidation indicators such as blood urea nitrogen (BUN), serum lactic acid (LD), lactic dehydrogenase (LDH), creatine kinase (CK), superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-Px), and reactive oxygen species (ROS) were measured by chemical colorimetry. The protein levels of pro-apoptotic protein B cell lymphoma-2 (Bcl-2)-associated X protein (Bax), anti-apoptotic Bcl-2 and cleaved cysteinyl aspartate-specific protease-3 (cleaved Caspase-3) in mouse skeletal muscle were detected by Western blot. ResultIn the rotarod test, the time on rod was shorter in the model group than in the blank control group (P<0.01) and the root, stem and fruit polysaccharide groups (P<0.01). In the forelimb grip strength test, the forelimb grip strength in the model group was lower than that in the blank control group (P<0.01) and the root, stem, leaf and fruit polysaccharide groups (P<0.01). In the weight-loaded swimming test, the weight-loaded swimming time in the model group was shorter than that in the blank control group (P<0.01) and the root, stem, leaf and fruit polysaccharide groups (P<0.01). Compared with those in the blank control group, the BUN, LD, LDH and CK levels significantly increased in the model group (P<0.05, P<0.01). The increases in BUN and LDH levels were decreased by the root, stem and fruit polysaccharides (P<0.05, P<0.01) and those in LD and CK by the root, stem, leaf and fruit polysaccharides (P<0.05, P<0.01). Compared with the blank control group, the model group showed decreased SOD and GSH-Px activities (P<0.01) and increased MDA and ROS content (P<0.01). Compared with the model group, the root, stem, and fruit polysaccharide increased the SOD activity (P<0.05, P<0.01) and decreased ROS content (P<0.01). The root and stem polysaccharides decreased the MDA content (P<0.01) and increased the GSH-Px activity (P<0.05, P<0.01). Compared with the blank control group, the model group showed up-regulated protein levels of Bax and cleaved Caspase-3 and down-regulated protein level of Bcl-2 (P<0.01). Compared with the model group, the root, and stem polysaccharides down-regulated the protein levels of cleaved Caspase-3 (P<0.05) and up-regulated protein level of Bcl-2 (P<0.01). ConclusionThe polysaccharides from the root, stem, leaf, and fruit of S. chinensis have anti-fatigue effect in D-galactose-induced aging mice. The polysaccharides may exert such effect by improving the antioxidant capacity and inhibiting the apoptosis of skeletal muscle cells.

ABSTRACT

OBJECTIVE@#To evaluate the efficacy of deep vein thrombosis (DVT) prevention among real-world surgical inpatients who received panax notoginseng saponins (PNS) combined with low-molecular-weight heparin (LMWH).@*METHODS@#A prospective cohort study was conducted among surgical patients between January 2016 and November 2018 in Xuanwu Hospital, Capital Medical University, Beijing, China. Participants received LMWH alone or PNS combined with LMWH for preventing DVT. The primary outcome was incidence of lower extremity DVT, which was screened once a week. Participants in the LMWH group were given LMWH (enoxaparin) via hypodermic injection, 4000-8000 AxalU once daily. Participants in the exposure group received PNS (Xuesaitong oral tablets, 100 mg, 3 times daily) combined with LMWH given the same as LMWH group.@*RESULTS@#Of the 325 patients screened for the study, 281 participants were included in the final analysis. The cohort was divided into PNS + LMWH group and LMWH group with 134 and 147 participants, respectively. There was a significant difference of DVT incidence between two groups (P=0.01), with 21 (15.7%) incident DVT in the PNS + LMWH group, and 41 (27.9%) incident DVT in the LMWH group. Compared with participants without DVT, the participants diagnosed with DVT were older and had higher D-dimer level. The multivariate logistic regression model showed a significant lower risk of incident DVT among participants in the PNS + LMWH group compared with the LMWH group (odds ratio 0.46, 95% confidence interval, 0.25-0.86). There were no significant differences in thromboelaslography values (including R, K, Angle, and MA) and differences in severe bleeding between two groups. No symptomatic pulmonary embolism occurred during the study.@*CONCLUSION@#Combined application of PNS and LMWH can effectively reduce the incidence of DVT among surgical inpatients compared with LMWH monotherapy, without increased risk of bleeding.

ABSTRACT

OBJECTIVES@#To explore the effect of polydatin on the proliferation and apoptosis of acute monocytic leukemia cell line THP-1 and the possible mechanism.@*METHODS@#After THP-1 cells were treated with polydatin at gradient concentrations for 24 hours and 48 hours, their proliferation was determined by CCK-8 assay, and half maximal inhibitory concentration (IC50) was calculated. Logarithmically growing THP-1 cells were divided into two groups, a polydatin treatment group (treated with IC50 of polydatin) and a blank control group (treated without polydatin solution), and incubated for 48 hours. Cell apoptosis and cell cycle were measured by flow cytometry. The expression levels of PI3K, AKT, p-AKT, mTOR, p-mTOR, p70 S6K, and p-p70 S6K proteins were measured by Western blotting.@*RESULTS@#After treatment with polydatin, the proliferation of THP-1 cells was strongly inhibited, and the IC50 at 48 hours was 1 800 μmol/L. After treatment with 1 800 μmol/L polydatin solution for 48 hours, the apoptosis rate of THP-1 cells increased significantly compared with the blank control group (P<0.05). The cell cycle was arrested in the G0/G1 and S phases, with a significantly increased proportion of cells in the G0/G1 phase and a significantly decreased proportion of cells in the S phase, as compared with the blank control group (P<0.05). The expression levels of PI3K, AKT, p-AKT, mTOR, p-mTOR, p70 S6K, and p-p70 S6K proteins decreased significantly compared with the blank control group (P<0.05).@*CONCLUSIONS@#Polydatin can effectively inhibit the proliferation, block the cell cycle, and induce the apoptosis of THP-1 cells, which may be related to inhibition of the PI3K/AKT/mTOR signaling pathway.

ABSTRACT

Excessive exercise makes the body consume more oxygen and produce excessive free radicals. The increased free radicals lead to oxidative stress injury and dysfunctions in liver tissue. Our previous study showed that Anwulignan, an active monomer in Schisandra sphenanthera Rehd. et Wils. (Schisandra), had anti-fatigue effects in mice. However, whether Anwulignan has a protective effect on liver damage in exhausted mice and the mechanism underlying remain elusive. An exhaustive swimming mice model was used to study the protective effects of Anwulignan on liver damage. The involvement of the nuclear factor (erythroid-derived 2)-like 2 (NRF2)/antioxidant responsive element (ARE) antioxidative pathway in Anwulignan-mediated anti-fatigue was analyzed using NRF2 inhibitor ML385 in HepG2 cells treated with H2O2. Animal welfare and experimental process follow the regulations of the Animal Ethics Committee of Beihua University. Anwulignan significantly lowered serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels, reduced liver tissue damages, increased superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT), and decreased malondialdehyde (MDA) and 8-hydroxy-2 deoxyguanosine (8-OHdG) contents in the livers of exhausted mice, demonstrating a strong antioxidant effect. Furthermore, Anwulignan up-regulated the NRF2/ARE antioxidant pathway in liver tissue, increased B-cell lymphoma 2 (Bcl-2) expression, and decreased Bcl-2-like protein 4 (Bax) and caspase3 expression. In HepG2 cells, Anwulignan improved the cell viability and SOD activity, reduced reactive oxygen species (ROS) and MDA contents, up-regulated the expression of the NRF2/ARE signaling pathway and Bcl-2, and decreased Bax and caspase3 expression in the cells. Furthermore, pretreated ML385 partly abolished all these effects of Anwulignan. Anwulignan protects the liver from damage in the exhausted mice by its antioxidant effects and related to its activation of the NRF2 pathway.

ABSTRACT

Elabelas/Toddlers belong to a group of endogenous active peptides recently discovered from zebrafish. The sequences of these peptides have 25% homology to Apelin. These peptides regulate physiological functions of organisms through putative protein receptors related to the angiotensin receptor AT1 (APJ). Functional roles of Elabela include early embryonic development, angiogenesis, fluid homeostasis, and feeding or dietary behavior control. Elabela also participates in the development of many diseases, such as heart failure, preeclampsia, acute kidney injury, hypertension, and diabetes. Increasingly, studies have shown that Elabela/APJ signaling can promote normal development of early embryos, including differentiation of mesoderm and endoderm, and formation of cardiac morphology and function. At the same time, the signaling can also promote angiogenesis or migration and proliferation of tumor cells. Here we describe the molecular structure, biological characteristics, functions and application prospects of Elabela/APJ signaling.

ABSTRACT

This study was designed to investigate the effect on tumor growth inhibition activity of lizards (Gekkoswinhonis Guenther) with different extent of broiling. Samples were prepared by a traditional drying method combined with broiling on clay tiles. Four groups of samples were all dried before broiling. Group A was without broiling; group B was mildly broiled; group C was moderately broiled; and group D was heavily broiled. Crispiness was detected by the sizes of the generated fragments of different groups and crispiness increased with broiling. Sensory evaluation of vision and olfaction was performed, and scores were generated by evaluators. Moderately broiled group had the highest total score in sensory evaluation. Water content and content of water-soluble extracts were detected according to Chinese Pharmacopoeia. With the increasing broiling extent, content of water-soluble extracts increased while water content decreased. Soluble protein concentration was detected by bicinchoninic acid (BCA) kit and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) with the same crude drug content. Soluble protein concentration decreased with the increasing broiling extent. With equal loading of proteins at the same concentration, soluble protein diversity was detected by SDS-PAGE. Band difference was marked by red boxes. Soluble protein molecule weights showed significant difference with the increasing broiling extent. H22 tumor-bearing mice model was established and used to detect tumor growth inhibition rate and immune organ index. Life quality of mice was evaluated. Mice treated with Gekkoswinhonis Guenther had better appetites and higher average weights compared with positive control group treated with fluorouracil (5-FU). Animal experiments were approved by the Ethics Committee of Beijing University of Chinese Medicine. Group A had the highest tumor growth inhibition rate (34.11%), followed by Group B (29.14%) and Group D (28.43%), Group C (21.98%) had the lowest tumor growth inhibition rate, but sensory evaluation was on the contrary. These results indicated that moderately broiling improved sensory evaluation but reduced the tumor growth inhibition activity of Gekkoswinhonis Guenther. The best tumor growth inhibition activity appeared when water content was 7.71%.

ABSTRACT

Objective To study the neuroprotective effect of orexin-B on rat model of cerebral ischemia-reperfusion injury and its molecular mechanism. Methods The artery occlusion model of male Wister rats(middle cerebral ar-tery occlusion,MCAO) was established which has been ischemic 2 h and reperfusion 24 h. Rats were randomly di-vided into sham group (control), ischemia-reperfusion group (I/R), ischemia-reperfusion +PBS group (I/R+PBS),and ischemia-reperfusion +orexin-B group (I/R+OXB). The neurological deficit scores were processed to inclusion and exclusion. Infarct size was determined by TTC staining;Using Western blot,the expressions of orexin receptor 2,p-AKT,p-GSK-3β proteins in hippocampus were detected;Jumping test was used to detect learning and memory abilities in rats. Results Orexin-B significantly reduced the volume of cerebral infarction in TTC staining;orexin-B group was significantly increased the expression of orexin receptor 2as well as p-AKT,which decreased p-GSK-3β (P<0.05),compared with the untreated group. Furthmore,the orexin-B treated group can improve the latency period and decline the mistakes in rat Jumping test(P<0.05). Conclusions The neuroprotective effect of orexin-B in cerebral ischemia-reperfusion injury may enhance p-AKT activity and inhibit p-GSK-3β activity,which may increase the proliferation of neurons and improve the cerebral blood glucose concentration.

ABSTRACT

Objective:To explore therapeutic effect of Xinmailong combined trimetazidine on patients with chronic heart failure (CHF).Methods:A total of 155 CHF inpatients hospitalized in our department from Oct 2013 to Apr 2016 were selected.According to therapeutic method,patients were divided into routine treatment group (n=50),Xinmailong group (n=52,received Xinmailong injection based on routine treatment group ) and combined treatment group (n=53,received trimetazidine based on Xinmailong group ),all groups were treated for eight weeks.Stroke volume (SV),cardiac output (CO),cardiac index (CI),left ventricular ejection fraction (LVEF),transmitral early diastolic peak flow velocity/late diastolic peak flow velocity (E/A),6min walking distance (6MWD) and total ef-fective rate were measured and compared among three groups before and after treatment.Results:Total effective rate of combined treatment group was significantly higher than that of routine treatment group (88.7% vs.66.0%,P=0.006),but there were no significant difference between combined treatment group and Xinmailong group (P=0.171).Compared with before treatment,after eight-week treatment,there were significant rise in SV,CO,CI,LVEF,E/A and 6MWD in three groups,P=0.001 all.Compared with routine treatment group after eight-week treatment,there were significant rise in SV,CI,LVEF and 6MWD in Xinmailong group,P<0.01 all;compared with routine treatment group and Xinmailong group,there were significant rise in SV [ (48.6 ± 8.9) ml vs.(53.6 ± 8.5) ml vs.(60.2 ± 8.4) ml],CO [ (4.4 ± 0.5) L/min vs.(4.5 ± 0.6) L/min vs.(5.7 ± 0.6) L/min],CI [ (3.1 ± 0.9) L·min-1·m-2vs.(3.7 ± 0.3) L·min-1·m-2vs.(4.3 ± 0.4) L·min-1·m-2],LVEF [ (42.3 ± 8.4)% vs.(46.6 ± 7.9)% vs.(53.2 ± 7.3)%],E/A [ (1.0 ± 0.3) vs.(1.1 ± 0.3) vs.(1.4 ± 0.2)] and 6MWD [ (342.88 ± 52.44) m vs.(420.90 ± 36.86) m vs.(443.55 ± 25.30) m] in combined treatment group,P=0.001 all.Conclusion:Xinmailong combined trimetazidine can improve cardiac function,hemodynamics,ventricular re-modeling and therapeutic effect in patients with chronic heart failure.

Objective Tolearntheparents'acceptanceofchargevaccinesandtheinfluencingfactors.Methods Totally 1869 parents with children of 0 -6 years old were selected with typical sampling and were investigated face to face.The awareness,acceptanceandinfluencingfactorsandservicedemandswereanalyzed.Results Therewere80.20%ofthe parents willing to give their children the charge vaccination.Compared with the local residents group,the other province resident group and other city resident group had a low probability to have charge vaccination (OR=0.777,OR=0.530). Parents who usually concerned about vaccine information were more likely to be vaccinated with charge vaccine (OR =1.307).Compared with other inoculation station,the stations which parents enjoying convenient service have higher inoculationrate(OR=1.673).Conclusion Mostparentshaveacceptedthechargevaccinationfortheirchildren. Health information and service quality of vaccination station are important influencing factors for accessibility of charge vaccination.

ABSTRACT

OBJECTIVE: To investigate the cognition status of construction workers on occupational health knowledge.METHODS: Seven hundreds and sixteen construction workers in Luzhou City were selected by cluster sampling method.Their cognition conditions on occupational health knowledge were investigated. RESULTS: The awareness rate of legal knowledge of occupational disease was the highest in construction workers in Luzhou City,which was 51. 68%. However,the awareness rate of legal compensation system of occupational disease was the lowest,which was only 5. 59%. The main sources of workers getting to know about occupational health knowledge were television, network and newspaper( accounting for 70. 25%). With the decrease of age and the increase of educational level,the awareness rates of occupational health knowledge in construction workers had a tendency of increase( P < 0. 05). The awareness rates of legal knowledge and diagnostic programs of occupational disease and legal compensation system in workers with special rates types of work were higher than those of handymen and traditional craftsmen( P < 0. 05). The awareness rates of occupational health knowledge in workers who had received the training of occupational safety and related occupational disease knowledge were higher than those who had not( P < 0. 05). The multivariate logistic regression analysis results showed that factors influencing the awareness rates of occupational health knowledge of construction workers were the educational level,type of work,the training of occupational safety and related occupational disease knowledge( P <0. 05). CONCLUSION: The awareness rate of occupational health knowledge in construction workers was generally low. The training of occupational safety and related occupational disease knowledge should be strengthened for the construction workers.

Objective To know the plasma lipids level and influencing factors among adults in Tongxiang city,and to provide evidence for prevention and intervention of dyslipidemia.Methods Multistage stratified cluster sampling method was conducted to select participants in Tongxiang city.Questionnaire interview,physical examination and blood lipid detection were performed among the selected subjects.Models of the univariate and multivariate unconditional logistic regressions were used to analyze the influencing factors of dyslipidemia.Results A total of 600 people were involved in this study.The prevalence and standardized rates of dyslipidemia were 36.33% and 36.11 %,and abnormal rates of HDL -C,TG,TC and LDL -C were 27.33%,15.67%,4.83% and 4.83%,which standardized were 29.17%,15.1 3%,3.1 8% and 3.12%, respectively.The abnormal rate of LDL -C in females was higher than that in males (P <0.05).Borderline abnormal rate of TC and dyslipidemia rates were increased with age (P <0.05),and the abnormal rate of HDL -C were decreased with age (P <0.05).Multivariate logistic regression analysis showed that BMI (OR =3.398),diabetes mellitus(OR =1.694), hypertension (OR =1.717)and total static time(OR =1.712)were the risk factors for dyslipidemia.Conclusion The prevalence of dyslipidemia was high among the adults in Tongxiang city,especially the abnormity of HDL -C and TG. Community comprehensive prevention and control of dyslipidemia should be carried out according to its risk factors.

Objective To know the influencing factors for the severity of motor vehicle road traffic injures ,and to provide scientific basis for making appropriate intervention measures.Methods 7 977 patients with motor vehicle road traffic injuries who were first diagnosed in injury surveillance sentinel hospital of Tongxiang City in 2012 and 2013 were selected, and the factors influencing the severity of injury were analyzed.Results The multivariate logistic regression analysis showed an increased risk of the injure severity among the people aged over 65 years old,alcohol drinking before the injury and migrant workers,the values of OR(95%CI)were 3. 231(2. 091 -4. 993),2. 796(1. 697-4. 607)and 1. 424(1. 217-1. 666)respectively.However,women,motor vehicle drivers,collision with non motor vehicle driver and using safety belt may contribute to the less serious damage,and the values of OR (95%CI)were 0. 837 (0. 753 -0. 930 ),0. 769 (0. 622-0. 952),0. 753 (0. 598-0. 948)and 0. 691 (0. 492-0. 970)respectively.Conclusion The risk factors for the severity of motor vehicle road traffic injures are elderly people,migrant workers and alcohol drinking.The safety education should be provided among the people and the targeted interventions should be developed and implemented.

ABSTRACT

<p><b>OBJECTIVE</b>To study the roles of follicular helper T (Tfh) cells and follicular regulatory T (Tfr) cells in the pathogenesis of Henoch-Schönlein purpura (HSP) in children.</p><p><b>METHODS</b>Peripheral blood samples were collected from 40 HSP children and 25 healthy controls. The percentages of Tfh and Tfr cells were measured by flow cytometry; the mRNA expression levels of Bcl-6, c-MAF, Blimp-1, and PD-1 in peripheral blood were measured by real-time polymerase chain reaction.</p><p><b>RESULTS</b>Compared with the controls, the children with HSP had significantly increased percentage of Tfh cells and Tfh/Tfr ratio but a significantly reduced percentage of Tfr cells in the peripheral blood (P<0.05). Compared with the controls, the children with HSP had significantly increased mRNA expression of Bcl-6 and c-MAF but significantly reduced mRNA expression of Blimp-1 in CD4+ T cells (P<0.05), and had significantly increased mRNA expression of PD-1 but significantly reduced mRNA expression of Blimp-1 in CD4+CD25+ regulatory T cells (P<0.05).</p><p><b>CONCLUSIONS</b>Abnormal percentages of Tfh and Tfr cells may be involved in the pathogenesis of HSP in children, and over-expression of Bcl-6, c-MAF, and PD-1 mRNA and inhibited expression of Blimp-1 mRNA may be considered as important reasons for abnormal percentages of Tfh and Tfr cells.</p>

Objective To exam the influence of psychological factors,occupational factors and dietary habits on malignant tumors.Methods A total of 2 471 patients with malignant tumor were selected from 2010 to 2012 and 1 to 1 matching method was used controlling sex and age distribution in the case and control groups.Data collected by interviewing was analyzed by univariate and multivariate conditional logistic regression.Results 12 related factors including 6 psychological factors were found in relation to malignant tumor after univariate logistic regression analysis.The multivariate logistic regression analysis showed that harmful occupational history (OR =1.731,95%CI:1.337 ~2.242),often having fried food (OR =1.889,95%CI:1.398 ~2.551),heavy salty diet(OR =1.457,95%CI:1.169 ~1.818),psychological problems (OR =1.274,95%CI:1.003 ~1.617),poor marital life (OR =2.007,95%CI:1.111 ~3.623)and lack of self -regulation (OR =1.460,95%CI:1.189 ~1.793 )were the risk factors.Conclusion Malignant tumor was related to a variety of exposure factors and psychological factor has become an important risk factor for malignant tumor in Tongxiang city.

ABSTRACT

Objective To study the ultrastructural features of Langerhans cell(LC),especially Birbeck granules in Langerhans cell histiocytosis(LCH) and provide some ultrastructural evidence to clarify the origin,function and stereoscopic structure of Birbeck granules.Methods Pathologic tissues of 8 LCH patients were treated with routine transmission electron microscope (TEM) procedures,fixture,dehydration,permeation,embedding,polymerization,ultramicrocut,staining and examined by TEM.Results Under the TEM,8 cases of LCH revealed typical LC cell,and LC cell in 6 cases revealed typical rod,tennnis-racquet like Birbeck granules.Some double membrane structures revealed irregular shape,sometimes accompanied with vacuole-like structure.Most of Birbeck granules locate inside the cytoplasmic membrane,in continuity with the cell membrane.There was Birbeck granule-like structure outside the cytoplasmic membrane in one case of multisystem LCH.Conclusions The ultrastructure of Birbeck granules could be various and irregular.The irregular Birbeck granule-like structure can also be useful for the diagnosis of LCH.Birbeck granules may arise from the cell membrane.The stereoscopic structure of Birbeck granules could be irregular.

ABSTRACT

<p><b>OBJECTIVE</b>To study the prognostic significance of CD47 in a NOD/SCID mouse model of acute myeloid leukemia (AML) and the best strategy for targeted therapy for this disease.</p><p><b>METHODS</b>CD34(+)CD38(-) leukemia stem cells (LSCs) were separated and transplanted into NOD/SCID mice to establish a mouse model of acute monocytic leukemia (AMoL). Anti-human CD47 antibody, alone or combined with cytosine arabinoside (Ara-C), was used to treat the mice with AMoL for 7-14 days, and therapeutic efficacy was assessed. LSCs were cultured together with mouse macrophages in culture medium containing anti-CD47 or anti-CD45 monoclonal antibody for 2 hours, to observe the phagocytic ability of macrophages to LSCs.</p><p><b>RESULTS</b>CD34(+)CD38(-) LSCs existed among THP-1 cells, with a content of about (0.12 ± 0.06)%, and a mouse model of AML was successfully established after the purified CD34(+)CD38(-) LSCs (97.0% ± 1.7%) were transplanted into NOD/SCID mice. The in vivo experiment showed that mice with AMoL had the most significant decrease in CD33(+)CD45(+) leukemia cells in peripheral blood and bone marrow and survived the longest after being treated with Ara-C (7 days) plus anti-CD47 monoclonal antibody (14 days) (P < 0.01). After 2 hours of in vitro culture, the phagocytic index in the culture medium containing anti-CD47 monoclonal antibody was significantly higher than in the culture medium containing anti-CD45 monoclonal antibody (76.9% ± 12.2% vs 7.60% ± 2.4%; P < 0.05).</p><p><b>CONCLUSIONS</b>High expression of CD47 is an adverse prognostic factor in AML. Combination therapy with anti-CD47 monoclonal antibody and Ara-C can effectively eliminate leukemia cells and LSCs, demonstrating great clinical significance in curing AML.</p>

ABSTRACT

A total of 24 biologically pure entophytic fungal strains were isolated from stems, leaves, and seed coats of Xylocarpus plants by repeated purification, and identified with Internal Transcribed Spacer (ITS) rDNA molecular method, which belonging to 14 genera, 11 families, 9 orders and 3 classes. There were differences in genus and species levels among three plant materials from different habitats and species, and it was found that the strains of Phomopsis and Colletotrichum existed in all three plant materials. In vitro assay of antitumor activity by MTT method revealed that the EtOAc extracts of 15 strains exhibited potent antitumor activity. These results suggest that it is of value for further investigation on the above fungal strains.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the role of Helicobacter pylori (Hp) and its products cytotoxin-associated protein (Cag A), vacuolating cytotoxin (VacA) in childhood acute idiopathic thrombocytopenic purpura (aITP), to evaluate the effect of Hp on their clinical outcome.</p><p><b>METHODS</b>Subjects were enrolled according to case-control design, including 184 aITP children and 154 healthy controls. They were inquired for demographic characteristics, the risk factors regarding Hp infection and ITP through a uniformed questionnaire. Patients with Hp infection were diagnosed by combined detection of serum Hp antibodies and stool antigens. CagA and VacA proteins were tested by ELISA method. In addition, clinical data and follow-up data of aITP children were collected. Non-conditional logistic regression and t test were applied for statistical analysis.</p><p><b>RESULTS</b>(1) The prevalence of Hp infection in aITP children and controls were 41.30% and 35.71%, respectively. No association between Hp infection and children aITP was found with OR of 1.170 (95%CI: 0.7163 - 1.673) after adjusting for confounding variables. (2) No statistical differences regarding initial platelet counts, megakaryocytes counts and the constituent ratio were found between the aITP children with and without Hp infection (P > 0.05). (3) No differences regarding initial platelet counts were found between aITP children with and without the expression of CagA (P > 0.05). The follow-up data showed that 32.88% of aITP children with Hp infection, as well as 29.70% of aITP children without Hp infection developed into cITP. No association between Hp infection and development to cITP was found with adjusted OR 1.171 (95%CI: 0.555 - 2.11 2).</p><p><b>CONCLUSIONS</b>The results didn't suggest that Hp is unlikely to play a role in the onset of childhood aITP, and in the development of aITP to cITP.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the concentration level and characteristics of indoor particle matter ≤ 2.5 µm in aerodynamic diameter (PM2.5) under extreme weather condition.</p><p><b>METHODS</b>During the period of haze in January 2013 and the fireworks and firecracker setting off in the Spring Festival of February 2013, three monitoring sites located indoor and outdoor were respectively selected by Beijing CDC, considering the vertical and horizontal distance, windows tightness and human activity indoor. PM2.5 samples were collected by filters and measured by laboratory weight method. At the same time, the meteorological data was also collected.</p><p><b>RESULTS</b>The median (quartiles) of overall concentration level of indoor PM2.5 was 87.76 µg/m(3) (52.05-174.48 µg/m(3)) and was lower than that of outdoor PM2.5 (128.79 µg/m(3), 95.14-221.88 µg/m(3); Z = -4.13, P < 0.01). The concentration of three indoor monitoring sites was different (χ(2) = 23.09, P < 0.01). The PM2.5 concentrations of monitoring point B in poor sealing window was the highest (94.05 µg/m(3); 63.46-189.17 µg/m(3)) and point C in sealed and less human activity, which was the lowest (77.89 µg/m(3), 51.19-144.40 µg/m(3)). The concentration level of indoor PM2.5 in the haze period (273.22 µg/m(3), 223.44-308.47 µg/m(3)) was higher than the overall concentration level of indoor PM2.5 (Z = -5.20, P < 0.01). The concentration level of indoor PM2.5 in the fireworks and firecracker period (167.90 µg/m(3), 129.15-187.90 µg/m(3)) was higher than that in the Spring Festival period (7 days, 72.76 µg/m(3), 36.97-145.30 µg/m(3), Z = -2.34, P < 0.05) and the overall concentration level of indoor PM2.5 (Z = -1.98, P < 0.05); however, it was lower than the concentration level of indoor PM2.5 in the haze period (Z = -3.43, P < 0.01). The I/O ratio (indoor concentration/outdoor concentration) was all less than 1.00 except 4, which was between 1.00-1.09. The mean I/O ratio in monitoring site B, monitoring site A and monitoring site C was 0.69 ± 0.21, 0.64 ± 0.23 and 0.58 ± 0.18, respectively, show significant bias (F = 22.85, P < 0.01). During the period of haze, the fireworks and firecracker and fine weather (when ambient PM2.5 concentration was lower than the standard value of 75 µg/m(3)), the mean I/O ratio was 0.87 ± 0.14, 0.68 ± 0.08 and 0.51 ± 0.18, respectively, showing significant bias (F = 29.88, P < 0.05). Under conditions of snow and high wind speed ( ≥ 3.4 m/s), PM2.5 concentration decreased to the valley point. The valley value of I/O ratio only occurred after several days of high windy weather. Moreover, the PM2.5 concentration level of indoor air showed a delayed 1-2 days after the haze weakened or disappeared.</p><p><b>CONCLUSION</b>Mass concentration of indoor PM2.5 increased significantly with increases of outdoor concentration. Haze and setting off fireworks/firecracker could lead to a serious decline of indoor air quality (IAQ), and the improvement of IAQ was lagging behind the outdoor changes.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effectiveness and safety of immunosuppressive therapy (IST) in the treatment of childhood aplastic anemia (AA) and to study the main factors influencing the effectiveness.</p><p><b>METHODS</b>The clinical data of 55 children with severe aplastic anemia (SAA) and 51 children with chronic aplastic anemia (CAA) were retrospectively analyzed. All patients received IST from January 2007 to December 2010.</p><p><b>RESULTS</b>In children with CAA, the effective rate of antithymocyte globulin (ATG) plus cyclosporine A(CsA) combination therapy was significantly higher than that of CsA alone (80% vs 44%; P<0.05); in children with SAA, the effective rate of ATG plus CsA combination therapy was also significantly higher than that of CsA alone (75% vs 40%; P<0.05). No patients developed clonal disease such as myelodysplastic syndrome, paroxysmal nocturn hemoglobinuria or acute myelocytic leukemia. In patients treated with the ATG plus CsA combination therapy, the response rate was relatively high for children whose disease course was less than six months, bone marrow hematopoietic area was more than 40%, had no severe infections, and experienced granulocyte colony stimulating factor (G-CSF) reaction during the early treatment; however, it was not related to AA subtypes and age.</p><p><b>CONCLUSIONS</b>ATG plus CsA combination therapy is effective and safe in the treatment of childhood AA. The disease course, bone marrow hematopoietic area, severe infections and G-CSF reaction to early treatment are the main factors influencing the therapeutic effects.</p>
